Experimental Study on the Effect of Hepatocyte Growth Factor for the Liver regeneration after Partial Hepatectomy


Abstract
Hepatocyte factor(HGF) is the most potent mitogen for mature hepatocyte and stimulates their DNA synthesis in experimental animals as well as humans. Author tried to asscess the effect of HGF on liver regeneration after 75% partial hepatectomy
(PH)
Healthy adult New Zealand white rabbits weighting about 2Kg were divided into three groups, sham operation group as control(n=5), 75%hepatctomy group (PH group:n=35), and HGF administration group after 75% PH (HGF group: n=35). The 5 rabbits
sacrificed
at 12 hours, 1 day, 7 days, 14 days, 21 days, and 28 days after PH in PH group and HGF group.
The liver function test, H-E stain, PAS stain and Ag-NOR stain were carried out in all groups, and the regeneration ratios were checked in the PH and HGF group to evaluate the effect of HGF after PH.
@ES The results obtained were summerized as follows.
@EN 1. The mean regeneration ratio of residual livers was 36.8% at 1 days, 54.7% at 3 days, 83.8% at 7 days, 105.4% at 14 days, 116.9% at 21 days, 116.2% at 28 days after PH: and In HGF group 36.9% at 1 day, 61.8% at 3 days, 90.4% at 7 days,
126.4%
at
14 days, 156.9% at 21 days, and 182.5% at 287 days after PH.
The regeneration ratio of HGF group was significantly higher than that of PH group at 14 days, 21 days and 28 days after PH.
2. The mitotic figures, double nucleated cells, and dark cells in PH group reached the peak level at 1~3 days and returned to control level at 14~21 days after PH. But the degree of mitotic figures and dark cells in HGF group was higher than in
PH
group.
3. The positive PAS reaction in PH group decreased significant from 12 hours to 7 days and was comparable to control level 4 days after PH.
But there were no difference between PH and HGF group in positive PAS reaction.
The mean value of Ag-NOR index (numbers of NORs/100 hepatocytes) was 170.2 in control group. In PH group, the mean value of Ag-NOR index was 218.4 at 1 day, and at 3 days and reached its peak, and returned to control level at 14 days after PH.
The mean value of Ag-NORs index in HGF group was 285.4 at 1 day and reached its peak and 264.6 at 3 days, 220.4 at 7 days and returned to control level at 21 days after PH.
The degree of Ag-NOR index in HGF group was higher and more persistant than in PH group
4. The mean value of AST and in PH group reached the peak level at 1 day after PH and returned to control level at 14 days after PH.
In HGF group, the mean have value of AST and ALT was significantly less than that in PH group and returned to control level at 14 days.
But there were no differences between PH and HGF group in ALP level.
